About
PT-141, also known as bremelanotide, is a cyclic heptapeptide analogue of alpha-melanocyte stimulating hormone. It was developed from work on melanotan II and holds an approved indication in some jurisdictions, which places it among the more formally characterised compounds in this catalogue.
Mechanism
The peptide is an agonist at melanocortin receptors, with activity described principally at the MC4 receptor in the central nervous system. This distinguishes its described mechanism from that of PDE5 inhibitors, which act on peripheral vascular smooth muscle: the melanocortin pathway is characterised in the literature as operating upstream, within central sexual-response circuitry. Reported effects on blood pressure are attributed to melanocortin activity in cardiovascular regulation.

Bremelanotide has been evaluated in randomised clinical trials for hypoactive sexual desire disorder in premenopausal women.
